USEFUL TIPS FOR FLOW CYTOMETRY

Panel Selection Tips

Note - Many cytometers are configured to minimize spillover effects and maximize success of your panel. Tips provided are general best practices.

Match BRIGHT colors with mid to low expressing markers.

Match DIM colors with mid to high expressing markers.

Avoid using dim colors with low expressing markers.

When possible, take advantage of multiple lasers and channels to keep spillover minimized by using channels not next to each other or close in emission range used.

Brightness of the Fluorochrome can affect spillover if the cell marker being labeled is abundant AND/OR close to another channel.

Choosing colors next to each other on the same laser can increase spectral overlap, especially if choosing 3 or more consecutive channels using bright colors.

Choosing tandem dyes for Intracellular Staining can impede staining quality (ex PECy5, PECy7, APC-Cy7).
