Sample Tubes Requirements

MoFlo XDP - 12x75mm (5mL) polystyrene or polypropylene round bottom tubes/0.5ml, 1.5ml eppendroff / 15 ml round/conical bottom tubes/ 50ml conical bottom tubes

Gallio's - 12x75mm (5ml) Polypropylene plastic tubes only.

Fortessa - 12X75mm (5ml) Polystyrene plastic tubes only.

Sort Collection Tubes

Use either 12x75mm (5mL) polypropylene plastic tubes or 12x120mm (15mL) polypropylene.

Note: Polystyrene tubes are not ideal as they accumulate charges during sorting which can result losses of sorted cells.

  • For the 12x75mm tubes place ~1ml of media in the tube.
  • For the 12x120mm tubes place ~2ml of media in the tube.
  • 1.5ml eppendroff with 0.3ml of media in the tube.

Sorting information

Running media:
1% FBS in any phenol-red-free media. HEPES can be added at a final concentration of 25mM to media to keep cells well buffered for all-day sorts, and/or to collection media.

Collection media:
this is usually 20% FBS in whichever media the cells are happiest. This is diluted 4-5x and ends up being approximately 5% FBS.

Collection plates:
Bring plates for collection with media pre-loaded in the wells. Do not fill wells above 1/2 maximum capacity to avoid spilling.

Instrument Clogs
Filter your samples with nylon mesh filters prior to running on any instrument.

Instrument Cleaning
Follow the cleaning listed in the Quick Reference or affixed to the instrument. Please clean any spills and throw any used, gloves, pipet tips, tube, tube caps, aluminum foil...in the trash.

Time Management
Make sure you stop your run before the end of your scheduled time to perform cleaning and in order not to delay the next user.

Food and Drinks
Food and Drinks are not allowed in the Flow Cytometry Facility.
